PT-141
PT-141 (Bremelanotide) is an FDA-approved synthetic peptide that acts as a melanocortin receptor agonist, specifically targeting MC3R and MC4R receptors in the central nervous system.
Also known as: Melanocortin Receptor Agonist, Sexual Dysfunction Treatment
Overview
PT-141 (Bremelanotide) is an FDA-approved synthetic peptide that acts as a melanocortin receptor agonist, specifically targeting MC3R and MC4R receptors in the central nervous system. Unlike traditional ED medications that work through vascular mechanisms, PT-141 directly influences sexual arousal pathways in the brain, making it effective for both male and female sexual dysfunction.
Key Benefits
- Hypoactive Sexual Desire Disorder (HSDD)
- Erectile Dysfunction
- Female Sexual Arousal Disorder
FDA-approved route with predictable absorption, works within 45 minutes, effective for both male and female sexual dysfunction
Mechanism of Action
Activates melanocortin receptors (MC3R/MC4R) in the central nervous system, triggering sexual arousal pathways independent of vascular mechanisms
Pharmacokinetics
Peak plasma concentration: 1 hr. Elimination half-life: 2.7 hrs. Largely cleared by: ~13.5 hrs.
Research Protocols Injectable
| Goal | Dose | Frequency | Route |
|---|---|---|---|
| Female HSDD (FDA-approved) | 1.75mg | As needed, max 1 dose/24hr | Subcutaneous |
| Male Erectile Dysfunction | 1-2mg | As needed, 45-60min before activity | Subcutaneous |
| Female Arousal Disorder | 0.75-1.25mg | As needed, max 1 dose/24hr | Subcutaneous |
| Low Starting Dose | 0.5mg | Test dose to assess tolerance | Subcutaneous |

Peptide Interactions
- Sildenafil/Tadalafil — Monitor Combination: Can be used together but monitor for additive blood pressure effects. PT-141 works centrally while PDE5 inhibitors work peripherally.
- Melanotan II — Use Caution: Both are melanocortin agonists - combining may lead to excessive melanocortin activation and side effects. Not recommended without medical supervision.
- Blood Pressure Medications — Monitor Combination: PT-141 can transiently lower blood pressure. Monitor BP closely when combining with antihypertensives.
- Alcohol — Use Caution: Both can lower blood pressure and cause flushing. Limit alcohol intake when using PT-141 to avoid excessive hypotension.
- Nitrates — Avoid Combination: Risk of severe hypotension. Do not use PT-141 with nitrate medications for heart conditions.
- BPC-157 — Compatible: No known interactions. Different mechanisms of action - PT-141 for sexual function, BPC-157 for tissue repair.
- Testosterone — Synergistic: May work synergistically for sexual dysfunction. PT-141 addresses arousal while testosterone supports libido and function.
- Kisspeptin — Monitor Combination: Both affect sexual function through different pathways - PT-141 via melanocortin receptors (MC3R/MC4R), Kisspeptin via GnRH stimulation. No receptor overlap but monitor for additive effects on blood pressure and hormonal response.
Peptide Instructions Injectable
Supplies:
- Bacteriostatic water (BAC water)
- Insulin syringes (29-31 gauge)
- Alcohol swabs
- PT-141 lyophilized powder vial
How to Reconstitute Injectable
- 1Allow vial to reach room temperature (15-20 minutes)
- 2Clean vial top with alcohol swab and allow to dry
- 3Calculate required bacteriostatic water volume using the calculator below
- 4Draw calculated volume of bacteriostatic water into syringe
- 5Inject water slowly down the inside wall of the vial (never directly onto powder)
- 6Gently swirl until powder completely dissolves (never shake)
- 7Solution should be clear - discard if cloudy or contains particles
- 8Store reconstituted solution in refrigerator at 2-8°C
What to Expect Injectable
First 30-45 minutes: May experience mild nausea or facial flushing. 45-90 minutes: Onset of effects - increased arousal and desire. 2-4 hours: Peak effects - enhanced sexual response and satisfaction. 6-12 hours: Effects gradually diminish. Long-term: No tolerance development reported with intermittent use.
Side Effects & Safety
Common side effects: nausea (40%), flushing (20%), headache (11%). Take anti-nausea medication 30 minutes before if prone to nausea. Monitor blood pressure - can cause transient decrease. Not for use with uncontrolled hypertension or cardiovascular disease. Avoid alcohol to minimize blood pressure effects. FDA black box warning for blood pressure effects.
